目的 研究SMMC - 772 1细胞诱导耐药培养中P -糖蛋白变化。方法 用MTT法检测常用化疗药物对SMMC - 772 1和SMMC - 772 1/ADM细胞的毒性 ;用流式细胞仪检测SMMC - 772 1/ADM细胞P -糖蛋白表达及细胞内柔红霉素的浓度。结果 随着培养液中阿霉素浓度的递增 ,SMMC- 772 1/ADM细胞内柔红霉素的积聚减少 ,P -糖蛋白表达有逐步增加的趋势。结论 P -糖蛋白的过度表达可能是形成SMMC - 772 1细胞多药耐药的重要原因。
Objective To study the changes of P - glycoprotein in SMMC - 772 1 cells induced by drug - resistant culture. Methods MTT assay was used to detect the cytotoxicity of commonly used chemotherapeutic drugs on SMMC - 772 1 and SMMC - 772 1 / ADM cells. The expression of P - glycoprotein and intracellular daunorubicin in SMMC - 772 1 / ADM cells were detected by flow cytometry concentration. Results With the increasing of doxorubicin concentration in culture medium, the accumulation of daunorubicin in SMMC-772 1 / ADM cells decreased and the expression of P - glycoprotein tended to increase gradually. Conclusion Overexpression of P - glycoprotein may be an important reason for multidrug resistance in SMMC - 772 1 cells.
