目的研究糖尿病患者皮肤组织中Ⅳ型胶原的糖基化对表皮干细胞生物学特性的影响。方法体外培养表皮干细胞,用免疫细胞化学和免疫荧光化学方法鉴定其标志物(角蛋白19和β1整合素),将表皮干细胞分别接种在包被有正常Ⅳ型胶原和糖基化Ⅳ型胶原的培养板上,比较两者黏附率、生长曲线和克隆生成率。结果糖基化Ⅳ型胶原上生长的表皮干细胞的黏附率和克隆生成率均较正常胶原组低(P<0.05)。结论Ⅳ型胶原的糖基化抑制了表皮干细胞的黏附与增殖,可能是糖尿病患者创面难愈的机制之一。
Objective To investigate the effect of glycosylation of type Ⅳ collagen on the biological characteristics of epidermal stem cells in the skin of diabetic patients. Methods Epidermal stem cells were cultured in vitro and their markers (cytokeratin 19 and β1 integrin) were identified by immunocytochemistry and immunofluorescence staining. Epidermal stem cells were inoculated into normal collagen Ⅳ and glycosylated collagen Ⅳ On the plate, the adhesion rate, growth curve and clonogenic rate were compared. Results The adhesion rate and clonogenic rate of epidermal stem cells grown on glycosylated type Ⅳ collagen were lower than those of normal collagen group (P <0.05). Conclusion Glycosylation of type Ⅳ collagen inhibits the adhesion and proliferation of epidermal stem cells, which may be one of the mechanisms of wound healing in diabetic patients.
