目的:研究复方中药芪苈强心胶囊的化学成分,从而明确芪苈强心胶囊的药效物质基础。方法:使用凝胶柱色谱、中低压液相色谱和制备高效液相色谱等手段,对芪苈强心胶囊大孔树脂50%和10%醇洗部分的化学成分进行分离和鉴定。结果:从这两段洗脱物中分离纯化得到12个单体化合物。利用1D,2D核磁共振光谱全部鉴定了其结构,分别为5’,7-二羟基-3’-甲氧基异黄酮(1),人参皂苷Rb1(2),人参皂苷Rf(3),人参皂苷Re(4),2’-羟基-3’,4’-二甲氧基异黄烷-7-O-β-D-葡萄糖苷(5),芒柄花素7-O-β-D-葡萄糖苷(6),(20R)-人参皂苷Rh1(7),8Z-decaene-4,6-diyne-1-O-β-D-glucopyranoside(8),杠柳毒苷(9),5-羟基-2-甲氧基-苯甲醇(10),腺苷(11),槲皮素-3-O-β-D-吡喃葡萄糖基-7-O-β-龙胆双糖苷(12)。结论:化合物1,5~12均为首次从芪苈强心胶囊中分离得到。其中化合物10为一个新的天然产物。
OBJECTIVE: To study the chemical constituents of Qili Qiangxin capsule in order to clarify the pharmacodynamic basis of Qili Qiangxin Capsule. Methods: The chemical constituents of 50% and 10% alcohol-washed fractions of Qiliqiangxin macroporous resin were separated and identified by gel column chromatography, medium-low pressure liquid chromatography and preparative high performance liquid chromatography. Results: Twelve monomeric compounds were isolated and purified from the two fractions. Their structures were identified by 1D and 2D NMR spectra. They were 5 ’, 7-dihydroxy-3’-methoxyisoflavone (1), ginsenoside Rb1 (2), ginsenoside Rf (3) Saponins Re (4), 2’-hydroxy-3 ’, 4’-dimethoxyisoflavine-7-O-β-D-glucoside (5) Glucoside (6), (20R) -ginsenoside Rh1 (7), 8Z-decaene-4,6-diyne-1-O- β -D- glucopyranoside (8) (10), adenosine (11), quercetin -3-O-β-D-glucopyranosyl-7-O-β-gentiobioside ). Conclusion: Compounds 1, 5 ~ 12 were isolated from Qili Qiangxin capsule for the first time. Among them, compound 10 is a new natural product.
