研究保肝颗粒对急性化学性肝损伤小鼠TNF-α和IL-1的影响。将105只昆明种小鼠按体质量随机分成7组,即正常组、模型对照组、护肝片组、克劳克组、保肝颗粒高、中、低剂量组,每组15只。小鼠适应性喂养1周后,分组灌胃,正常组和模型组每天ig给予生理盐水,护肝片组给药量为2片·kg-1·d-1,克劳克组为25m L·kg-1·d-1,保肝颗粒高、中、低分别为12.9、6.45、3.225 g·kg-1·d-1,每天1次,连续30 d。末次给药1 h后,除空白对照组外,每组分别ip 0.2%CCl4油溶液0.01 m L·10 g-1,禁食不禁水24 h后,处死小鼠取材用以各项指标检测。采用双抗体夹心ELISA法测定实验小鼠血清及肝组织TNF-α、IL-1。结果表明,保肝颗粒能够改善CCl4所致急性化学性肝损伤小鼠肝细胞组织形态与结构,显著降低急性肝损伤小鼠血清及肝组织TNF-α、IL-1蛋白含量,其发生机理与调控细胞因子有关。
To study the effect of Baogan granule on TNF-α and IL-1 in mice with acute chemical liver injury. 105 Kunming mice were randomly divided into 7 groups according to body weight, namely normal group, model control group, Hugan tablet group, Clogroup, Baogu particles high, medium and low dose group, 15 rats in each group. One week after adaptive feeding, the mice were intragastrically gavaged with normal saline. Normal saline was given to the normal and model groups, respectively. The dosage of Hugan tablet was 2 tablets · kg-1 · d-1, · Kg-1 · d-1, Baogan Granules were 12.9,6.45 and 3.225 g · kg-1 · d-1, respectively, once a day for 30 days. At 1 h after the last administration, except for the blank control group, the rats in each group were treated with 0.2% CCl4 oil solution 0.01 m L · 10 g-1 for 24 h. The serum and liver tissue TNF-α and IL-1 in experimental mice were determined by double antibody sandwich ELISA. The results showed that Baogan Granule could improve the morphology and structure of liver cells in acute liver injury induced by CCl4 and significantly decrease the levels of TNF-α and IL-1 in serum and liver of acute liver injury mice Regulation of cytokines.
