目的探讨微小RNA-21(miR-21)对乳腺癌细胞MCF-7增殖的影响。方法实时荧光定量PCR(qRT-PCR)检测15例乳腺癌患者癌组织和癌旁组织中miR-21的表达。乳腺癌细胞株MCF-7用miR-21抑制物转染(抑制组,MCF-7+INH)、抑制物对照转染(对照组,MCF-7+INH-NC)和未转染(空白对照组)后,MTT法检测细胞增殖,流式细胞术分析细胞周期,Western blot检测程序性细胞凋亡因子4(PDCD4)表达。结果 miR-21在乳腺癌组织中高表达(P<0.05)。与对照组相比,miR-21抑制组MCF-7细胞的增殖抑制,G0/G1期细胞比例增多,S期细胞比例减少,PDCD4表达增加(P<0.05)。结论抑制乳腺癌细胞MCF-7中miR-21的表达,可以抑制肿瘤细胞的增殖。
Objective To investigate the effect of microRNA-21 (miR-21) on the proliferation of breast cancer cell line MCF-7. Methods Real-time quantitative PCR (qRT-PCR) was used to detect the expression of miR-21 in 15 cases of breast cancer and adjacent tissues. The breast cancer cell line MCF-7 was transfected with miR-21 inhibitor (MCF-7 + INH inhibition group), inhibitor control (MCF-7 + INH-NC) and untransfected Group). The cell proliferation was detected by MTT assay. The cell cycle was analyzed by flow cytometry. The expression of PDCD4 protein was detected by Western blot. Results miR-21 was highly expressed in breast cancer tissues (P <0.05). Compared with the control group, the proliferation of MCF-7 cells was inhibited in miR-21 group, the proportion of cells in G0 / G1 phase increased, the proportion of S phase cells decreased, and the expression of PDCD4 increased (P <0.05). Conclusions Inhibition of miR-21 expression in breast cancer cells MCF-7 can inhibit the proliferation of tumor cells.
