目的 探讨同种异系抗原经门静脉接种后机体对该抗原免疫反应状态的改变。方法 以NIH/q小鼠为供体 ,BALB/C小鼠为受体。预先用NIH/q小鼠脾细胞或与供受体无关C5 7/BL小鼠脾细胞注射至BALB/C小鼠门静脉或下腔静脉 ,1周后用NIH/q小鼠脾细胞注射至BALB/C小鼠背部皮下免疫 ,2周后第 2次皮下免疫。第 2次皮下接种后 1周 ,测定供受体之间的混合淋巴细胞反应 (MLR)和受体对供体抗原的迟发性超敏反应 (DTH)。结果 供体脾细胞门静脉接种组供受体混合淋巴细胞反应 (MLR)其刺激指数 (SI)为 3.5 1± 0 .5 5 ,DTH脚掌厚度增值为 (7.41± 0 .6 )× 10 -2mm ,而供体脾细胞下腔静脉接种组以及与供受体无关的C5 7/BL小鼠脾细胞门静脉接种组供受体MLR(SI)分别为 34.83± 3.90和 35 .33± 2 .5 4,DTH脚掌厚度增值分别为 (2 3 .5 1± 1.6 9)× 10 -2 mm和 (2 3 .87± 2 .76 )× 10 -2 mm ,差异有显著性 (P <0 .0 5 )。结论 同种异系抗原门静脉接种可诱导机体产生对该抗原的特异性免疫无反应 ,其作用具有抗原特异性。
Objective To investigate the change of the immune response of the alloantigen to the antigen after allogeneic antigen was inoculated through the portal vein. Methods NIH / q mice as donors and BALB / C mice as recipients. BALB / C mice were injected into the portal vein or inferior vena cava with NIH / q mouse splenocytes or splenocytes unrelated to donor recipients in advance and splenocytes of NIH / q mice were injected into the BALB after 1 week / C mice were subcutaneously immunized on the back, and 2 times subcutaneously after 2 weeks. One week after the second subcutaneous inoculation, the mixed lymphocyte reaction (MLR) between the donors and the delayed hypersensitivity (DTH) of the donor to the donor antigen were determined. Results The stimulating index (SI) of donor lymphocyte reaction (MLR) in portal vein of donor spleen cells was 3.5 1 ± 0.55, and the increment of the thickness of the sole of DTH was (7.41 ± 0. 6) × 10 -2mm, However, the MLR (SI) of donor spleen cells inoculated into the inferior vena cava and the portal vein inoculated with C5 7 / BL mice unrelated to donor were 34.83 ± 3.90 and 35.33 ± 2.54, respectively, The thicknesses of the soles of DTH were (21.51 ± 1.6 9) × 10 -2 mm and (23.87 ± 2.76) × 10 -2 mm respectively, with significant difference (P <0.05) . Conclusion Allogeneic antigenic portal vein inoculation can induce the body to produce specific immunity to this antigen without reaction, its function is antigen-specific.
