目的:探讨补阳还五汤含药血清对缺氧损伤PC12细胞的保护作用。方法:应用中药复方血清药理学研究方法,制备家兔补阳还五汤含药血清,观察其对缺氧PC12细胞损伤模型细胞形态学、存活力及上清液中PAF含量的影响。结果:PC12细胞缺氧造模后,出现急性损伤,形态变异,补阳还五汤含药血清组表现为自我修复,形态与正常细胞组相似;补阳还五汤含药血清组能提高细胞存活力,降低凋亡率及降低细胞上清PAF含量(P<0.05)。结论:补阳还五汤含药血清对缺氧损伤PC12细胞有明显的保护作用,其机制与降低缺氧损伤细胞PAF含量释放、保护细胞形态和提高存活力有关。
Objective: To investigate the protective effect of Buyang Huanwu Decoction-containing serum on hypoxic injury PC12 cells. Methods: Serum pharmacology of Chinese herbal compound recipe was used to prepare the medicine serum of Buyang Huanwu Decoction in rabbits. The morphological changes and viability of hypoxic PC12 cells and PAF content in supernatant were observed. Results: After hypoxia, PC12 cells showed acute injury and morphological changes. The drug-containing serum of Buyang Huanwu Decoction group showed self-repair and its morphology was similar to that of normal cells. The drug-containing serum of Buyang Huanwu Decoction could increase the number of cells The viability, the apoptosis rate and the content of PAF in the cell supernatant were decreased (P <0.05). Conclusion: The serum of Buyang Huanwu Decoction contains significant protective effect on hypoxic injury of PC12 cells, and its mechanism is related to reducing the release of PAF in hypoxic injury cells, protecting the cell morphology and improving the viability.
