目的探讨表皮生长因子(EGF),成纤维细胞生长因子(FGF),对人绒毛滋养层细胞体外生长的调控作用。方法对12例人早孕绒毛离体组织及16例复发性流产绒毛进行体外分离培养,细胞汇合后消化,做细胞增殖试验,在相同的生长期加入EGF,FGF,EGF+FGF培养24,48,72h,用MTT法、DNA切口末端标记法测定EGF,FCF对2组绒毛滋养层细胞的作用。结果 10.0ng/ml EGF及FGF刺激细胞生长,EGF+FGF刺激细胞生长的作用最强,10.0ng/ml EGF,FCF不刺激细胞凋亡,两组结果具有一致性,MTT法、DNA切口末端标记法结果一致。结论人滋养层细胞的生长受到EGF,FGF的调控,EGF及FGF具有促进细胞增殖作用;EGF,FGF联合应用具有最佳的协同效应。
Objective To investigate the regulatory effect of epidermal growth factor (EGF) and fibroblast growth factor (FGF) on the growth of human chorionic trophoblast cells in vitro. Methods 12 cases of early human villi in vitro and 16 cases of recurrent spontaneous abortion were isolated and cultured in vitro, the cells confluent digestion, cell proliferation test, in the same growth phase by adding EGF, FGF, EGF + FGF cultured 24,48, 72h. The effects of EGF and FCF on the two trophoblast cells were determined by MTT assay and DNA nick end labeling. Results EGF and FGF stimulated the cell growth at the concentrations of 10.0ng / ml and EGF. However, EGF and FGF did not stimulate the cell apoptosis. The results of the two groups were consistent. MTT and DNA nick end labeling Law consistent. Conclusion The growth of human trophoblast cells is regulated by EGF and FGF, while EGF and FGF promote cell proliferation. The combination of EGF and FGF has the best synergistic effect.
