目的:探讨抵当培元汤与5-Fu联合治疗移植性大肠癌BALB/c小鼠的疗效及作用机制。方法:BALB/c小鼠50只,雌雄各半。将荷瘤小鼠随机分为5组,除正常组之外,其余4组(模型对照组、抵当培元汤组、5-FU组、抵当培元汤+5-FU组)于小鼠左侧胁腹部皮下接种对数生长期的HCT-8细胞,制作移植性大肠癌模型。给药4周后,观察各组抑瘤率、免疫功能及氧自由基的清除能力的变化。结果:联合治疗组小鼠抑瘤率明显高于其它治疗各组;在胸腺指数、脾指数及T淋巴细胞增殖能力方面,抵当培元汤组好于5-FU组及联合用药组;三个药物治疗组及模型对照组较正常对照组的SOD和GSH—PX活性降低,而MDA含量增高,但三个药物治疗组明显好于模型对照组(P<0.05),联合治疗组的数据表现好于其它治疗组。结论:抵当培元汤能够改善HCT-8荷瘤小鼠免疫力低下和免疫紊乱状态,联合用药具有更好的抑制肿瘤及清除氧自由基的能力。
Objective: To investigate the curative effect and mechanism of combination of Dadang Peiyuan Decoction and 5-Fu on BALB / c mice. Methods: 50 BALB / c mice, half male and half female. Tumor-bearing mice were randomly divided into 5 groups. Except for the normal group, the other 4 groups (model control group, Weide Peiyuan Decoction group, 5-FU group, and Weide Peiyuan Decoction + 5-FU group) Hypothyroidism was subcutaneously inoculated with logarithmic growth phase HCT-8 cells to make a transplantable colorectal cancer model. After 4 weeks of administration, the inhibition rate of tumor, immune function and the scavenging ability of oxygen free radical in each group were observed. Results: The inhibitory rate of mice in the combined treatment group was significantly higher than that of other treatment groups; in the aspects of thymus index, spleen index and T lymphocyte proliferation ability, Compared with the normal control group, the activity of SOD and GSH-PX in the drug treatment group and the model control group decreased and the MDA content increased, but the three drug treatment groups were significantly better than the model control group (P <0.05), and the data of the combination treatment group showed good performance In other treatment groups. Conclusion: Doudang Peiyuan Decoction can improve the immunodeficiency and immune disorders in HCT-8 mice, and the combination therapy can better inhibit tumor and scavenge oxygen free radicals.
