应用大白鼠连续ig给药4周,对欧达胃克的长期用药毒性进行观察,结果,低剂量组(350mg·kg~(-1);相当于临床量的2.3倍)和中剂量组(700mg·kg~(-1),4.5倍量)与对照组比较各项指标无显著差异。高剂量组(1232.0mg·kg~(-1),8倍量)用药2周活动逐渐减少,饮食量下降,体重减轻。用药3周高剂量组有30%大鼠站立行走不便,出现肌张力障碍。测大鼠坐骨神经复合动作电位,表现抑制现象,兴奋阈降低。高剂量组BUN有可逆性升高。实验结束时有3只大鼠死于呼吸抑制(死亡率未超过20%)。
The long-term toxicity of Ondanweike was observed in rats using ig administration for 4 weeks.
