目的探讨吸烟对大鼠血清CD3、CD4、CD8 T淋巴细胞含量影响的时间效应。方法雄性健康清洁级Wistar大鼠30只,随机分为对照组和烟草烟雾暴露组,每组15只;对照组暴露于正常饲养环境,烟草烟雾暴露组每日暴露于烟草烟雾环境30 min,每日2次;2组大鼠分别于烟草烟雾暴露时(T0)、烟草烟雾暴露1个月(T1)、烟草烟雾暴露2个月(T2)、烟草烟雾暴露3个月(T3)、烟草烟雾暴露4个月(T4)、烟草烟雾暴露5个月(T5)和烟草烟雾暴露6个月(T6)时采內眦静脉血,检测血清CD3、CD4、CD8 T淋巴细胞含量。结果 T0时,烟草烟雾暴露组外周血CD3、CD4、CD8 T淋巴细胞含量分别为(52.68±4.64)%、(34.39±3.03)%、(29.74±2.62)%;烟草烟雾暴露组与对照组比较,T3~T6时CD3和CD4 T淋巴细胞含量均下降(均P<0.05),CD8 T淋巴细胞含量均增多(均P<0.05);各时点间比较,烟草烟雾暴露组CD3、CD4 T淋巴细胞自T3时逐渐下降,CD8 T淋巴细胞自T4时逐渐上升(均P<0.05),对照组各指标各时点间差异均无统计学意义。结论烟草烟雾暴露损伤细胞免疫的时间窗为3个月。
Objective To investigate the time effect of smoking on the serum levels of CD3, CD4 and CD8 T lymphocytes in rats. Methods Thirty male Wistar rats were randomly divided into control group and tobacco smoke exposure group, 15 rats in each group. The control group was exposed to normal feeding environment. The tobacco smoke exposure group was exposed to tobacco smoke for 30 min every day. Day two times. The two groups of rats were exposed to tobacco smoke (T0), tobacco smoke exposure for one month (T1), tobacco smoke exposure for two months (T2), tobacco smoke exposure for three months (T3) The blood samples were collected for 4 months (T4), 5 months of exposure to tobacco smoke (T5) and 6 months of exposure to tobacco smoke (T6). Serum levels of CD3, CD4 and CD8 T lymphocytes were measured. Results At T0, the levels of CD3, CD4 and CD8 T lymphocytes in peripheral blood of tobacco smoke exposure group were (52.68 ± 4.64)% and (34.39 ± 3.03)%, respectively (29.74 ± 2.62)%, compared with the control group CD3 and CD4 T lymphocytes were decreased (all P <0.05) and CD8 T lymphocytes were increased at T3 ~ T6 (all P <0.05) The number of CD8 T lymphocytes gradually decreased from T3 (all P <0.05). There was no significant difference in each index of control group at all time points. Conclusion The time window of cellular immunity of tobacco smoke exposure injury is 3 months.
