目的:探讨加味真武汤对肾间质纤维化大鼠转化生长因子(TGF-β)及纤维连接蛋白(FN)表达的影响。方法:大鼠随机分为假手术组、模型组及治疗组,采用单侧输尿管梗阻(UUO)模型,术后7,14,21 d分别处死大鼠,测定肾脏指数、血肌酐(Cr)、尿素氮(BUN);肾组织HE及Masson染色,观察肾脏病变;免疫组化法比较各组TGF-β及FN的表达。结果:术后随时间的延长,模型组和治疗组大鼠可见明显的肾间质纤维化改变,肾间质TGF-β,FN的表达明显高于假手术组(P<0.05)。治疗组大鼠肾间质纤维化程度及TGF-β,FN的表达均低于模型组(P<0.05)。结论:加味真武汤可降低UUO模型大鼠血Cr,BUN水平,抑制TGF-β表达,减少FN形成,对肾间质纤维化有积极的治疗意义。
Objective: To investigate the effect of Jiawei Zhenwu Decoction on the expression of transforming growth factor (TGF-β) and fibronectin (FN) in rats with renal interstitial fibrosis. Methods: The rats were randomly divided into sham operation group, model group and treatment group. Unilateral ureteral obstruction (UUO) model was used. Rats were sacrificed on the 7th, 14th and 21st day after operation. The renal index, serum creatinine Urea nitrogen (BUN), renal tissue HE and Masson staining were used to observe the renal pathological changes. The expression of TGF-β and FN in each group was compared by immunohistochemistry. Results: With the extension of time, obvious changes of renal interstitial fibrosis were observed in model group and treatment group. The expression of TGF-β and FN in renal interstitium was significantly higher than that in sham operation group (P <0.05). The level of renal interstitial fibrosis and the expression of TGF-β and FN in treatment group were lower than those in model group (P <0.05). Conclusion: Jiawei Zhenwu Decoction can reduce the level of blood Cr and BUN, inhibit the expression of TGF-β, reduce the formation of FN in UUO rats, and have a positive therapeutic effect on renal interstitial fibrosis.
