近年来随着对树木品种改良工作的深入研究,极需各种树木染色体的资料。国外曾对日本柳杉(Cryptomeria japonica D.DON)的染色体作了一些研究,本文就中国柳杉(Cryptomera fortunei Hooibrenk ex Otto et Dietr.)的染色体研究并与国外日本柳杉染色体研究报道作比较分析,现报道如下: 材料和方法 (一) 取材:材料采自本校校园内中国柳杉树上生长的种子,在25℃恒温下培养。 (二) 预处理:将萌发后的种子置于饱和对二氯苯水溶液中处理12小时。 (三) 压片染色镜检:用改良的石碳酸品红染色30分钟左右,镜检500个制片,对其中分散较好的100个细胞作了染色体计数。
In recent years, with the in-depth study of the improvement of tree species, very much need for a variety of tree chromosome information. Some foreign countries have done some research on chromosomes of Cryptomeria japonica D.DON. In this paper, the chromosome of Cryptomeria fortunei Hooibrenk ex Otto et Dietr. Was studied and compared with the reports of chromosomes of Cryptomeria japonica , Are reported as follows: Materials and methods (a) drawn: Materials collected from the campus of Chinese cedar trees grown seeds, cultured at 25 ℃ constant temperature. (B) Pretreatment: Seeds germinated after germination of dichlorobenzene aqueous solution for 12 hours. (C) Tableting Dyeing Microscopy: Improved carbonate carmine magenta staining for about 30 minutes, microscopically examining 500 films and counting the chromosomes of well dispersed 100 cells.
