200g左右成年豚鼠17头分成对照及模型二组。模型组用25%d一半乳糖125mg/kg作球后注射,每天一次,连续34天。在第10天后作裂隙灯检查,晶体发现有空泡,20天后有晶体周边空泡及混浊,第34天摘眼球每鼠取1晶体作扫描电镜观察。制样方法:将晶体放入4%戊二醛磷酸缓冲液(1/15M,pH7.3)数日,1%锇酸后固定,丙酮梯度脱水,临界点干燥,沿前后轴断裂样品。
About 200 g of adult guinea pigs were divided into control and model 2 groups. The model group was injected intraperitoneally with 25% d of lactose 125 mg/kg once daily for 34 days. After the 10th day, a slit lamp examination was performed. The crystals were found to have vacuoles. After 20 days, there were vacuoles and turbidity around the lens. On the 34th day, the eyeball was taken from each mouse and a crystal was taken for scanning electron microscopy. Sample Preparation: The crystals were placed in 4% glutaraldehyde phosphate buffer (1/15M, pH 7.3) for several days, fixed with 1% citric acid, dehydrated with acetone gradient, dried at the critical point, and fractured along the anterior-posterior axis.
