目的 了解凋亡相关基因 p53、bcl 2、bax、Fas在鼻咽癌 (NPC)中的表达及相互关系。方法 在 67例NPC、5例癌旁组织中用免疫组化技术标记bcl 2、p53、bax、Fas ;用免疫双标技术同时标记 p53与bcl 2 ;从中选取 15例NPC用DNA原位杂交技术检测bcl 2基因扩增情况。结果 ① 67例NPC中 p53蛋白阳性 38例 (56 7% ) ,bcl 2蛋白阳性 4 8例(71 6% ) ,bax蛋白阳性 2 8例 (4 1 8% ) ,Fas蛋白阳性 2 9例 (4 3 3% )。②NPC中 ,p53蛋白与bcl 2蛋白的阳性率显著高于癌旁组织 ,而bax蛋白与Fas蛋白的阳性率却显著低于癌旁组织。③ p53蛋白与bcl 2蛋白阳性表达间呈正相关。④bcl 2 /bcl 2同二聚体在NPC中占优势 ;p53蛋白表达在bcl 2 /bcl 2同二聚体占优势的肿瘤中显著高于bax/bax同二聚体占优势的肿瘤中。结论 ①p53、bcl 2、bax、Fas在NPC中有较高的阳性表达率 ,与NPC的发生、发展关系密切。②NPC中 p53与bcl 2蛋白表达呈正相关 ,提示NPC中 p53蛋白主要不是突变型 ,有异于其它肿瘤。③抑制细胞凋亡的bcl 2 /bcl 2同二聚体在NPC中占优势 ,尤其是泡状核细胞癌中。提示其与NPC的发生及分化有关。
Objective To investigate the expression of p53, bcl 2, bax and Fas in nasopharyngeal carcinoma (NPC) and their correlations. Methods The bcl 2, p53, bax and Fas were labeled by immunohistochemistry in 67 cases of NPC and 5 cases of paracancerous tissues. P53 and bcl 2 were simultaneously labeled by double immunolabeling. Fifteen NPCs were selected by DNA in situ hybridization Detection of bcl 2 gene amplification. Results ① The 67 cases of p53 were positive in 38 cases (56.7%), 48 cases (71.6%) were positive for bcl 2 protein, 28 cases were positive for bax protein (41.8%), 29 cases were Fas protein positive 4 3 3%). ② The positive rate of p53 protein and bcl 2 protein in NPC was significantly higher than that in paracancerous tissues, while the positive rates of bax protein and Fas protein were significantly lower than those in paracancerous tissues. P53 protein and bcl 2 protein positive expression was positively correlated. Bcl2 / bcl2 homodimers predominate in NPC; p53 protein expression was significantly higher in tumors dominated by homodimers of bcl2 / bcl2 than in tumors predominated by homodimers of bax / bax. Conclusion ① The positive expression rate of p53, bcl 2, bax and Fas in NPC is high, which is closely related to the occurrence and development of NPC. ②NPC p53 and bcl 2 protein expression was positively correlated, suggesting that NPC in p53 protein is not the main mutant, different from other tumors. ③ Inhibition of apoptosis of bcl 2 / bcl 2 homodimers predominates in NPC, especially in somatic cell carcinomas. Suggesting that it is related to the occurrence and differentiation of NPC.
