[目的]建立同属不同种大黄土大黄苷测定方法。[方法]色谱柱C18柱(Agilent TC-C18 analytical,4.6mm×250mm,5μm),流动相乙腈-水(25:75),柱温30.0℃,检测波长323nm,流速0.80mL·min~(-1)。用DAD检测器200~400nm对土大黄苷对照品和供试品色谱进行光谱扫描。[结果]药用大黄中未见土大黄苷色谱峰,河套大黄和华北大黄均可见土大黄苷色谱峰。提示正品大黄中无土大黄苷,伪品大黄中含有土大黄苷。土大黄苷对照品和供试品出现一致色谱峰,结果显示二峰的光谱扫描图一致。提示供试品含有土大黄苷。[结论]薄层色谱法和高效液相色谱法检测结果准确,可做为鉴别大黄药材的推荐方法。
[Objective] The research aimed to establish a method for the determination of eosin in different kinds of loess. [Method] The mobile phase consisted of acetonitrile - water (25:75) with a column temperature of 30.0 ℃ and a detection wavelength of 323 nm at a flow rate of 0.80 mL · min -1 on a C18 column (Agilent TC-C18 analytical, 4.6 mm × 250 mm, 1). DAD detector with 200 ~ 400nm on the euphorbia glycosides reference substance and the chromatographic sample for spectral scanning. [Result] Rhubarb Rhubarb medicinal rhubarb rhizoma rhizome is not seen in chromatographic peaks, rhododendron Rhubarb and North Rhubarb are visible soil rhubarb glycosides peak. Tip genuine rhubarb soilless rhubarb glycosides, pseudo-rhubarb contains soil rhubarb glycosides. Soil emodin glycosides reference substance and the test sample consistent peaks, the results show that the second peak spectral scan consistent. Tips for the test substance containing emodin glycosides. [Conclusion] The results of TLC and HPLC are accurate and can be used as the recommended method to identify rhubarb herbs.
