用 SP_2/0小鼠骨髓瘤细胞与 EL-Tor 霍乱弧菌免疫的 Balb/c 小鼠脾细胞,在 PEG-1000作用下进行融合,获得9株分泌抗霍乱弧菌 McAb 的杂交瘤细胞系。用 ELISA 法对其中两株分泌的 McAb 进行特异性、敏感性实验,免疫球蛋白的类型鉴定及染色体检查。并用羟基磷灰石梯度提纯的 McAb 与兰州生研所生产的多克隆抗体(PcAb),对200例临床粪便标本,用双盲法进行快速检测。结果表明,McAb 的敏感性和特异性均优于 PcAb,同时也证实 McAb 应用于霍乱病人的诊断及菌株鉴定的可行性。
Spleen cells of Balb / c mice immunized with SP_2 / 0 mouse myeloma cells and Vibrio cholerae EL-Tor were fused by PEG-1000 to obtain 9 hybridoma cell lines secreting anti-Vibrio cholerae McAbs. The specificity and sensitivity of two McAbs secreted by ELISA were tested by ELISA. The types of immunoglobulins and chromosome examination were also studied. The McAb purified by hydroxyapatite gradient and polyclonal antibody (PcAb) produced by Lanzhou Institute of Health were used for rapid detection of 200 clinical stool specimens by double-blind method. The results showed that McAb sensitivity and specificity are better than PcAb, also confirmed the application of McAb in the diagnosis of cholera patients and the feasibility of identification of strains.
