目的:探讨沙棘籽粕中蛋白质提取工艺研究。方法:以蛋白质含量为24.97%的沙棘籽粕为原料,对沙棘籽粕进行粉碎,得到过80目标准药筛的沙棘粉末。将沙棘粉末进行醇提,其最佳工艺如下:乙醇浓度75%、料液比1∶6、提取温度50℃、提取3次每次60 min。然后用碱提酸沉法及双氧水脱色法对醇提沙棘蛋白进行精制,其工艺参数为:pH=10、料液比1∶12、碱提温度50℃、碱提3次每次50 min、30%双氧水与碱提液比例1∶20、脱色温度45℃、脱色时间60 min,酸沉pH=5。结果:经过粉碎过筛、醇、碱及脱色三步法提取后,得到了沙棘蛋白产品,其蛋白质纯度为84.18%,产品得率为11.39%。结论:该提取工艺效果更好。
Objective: To study the protein extraction process of sea buckthorn seed meal. Methods: Seabuckthorn seed meal with protein content of 24.97% was used as raw material to pulverize sea buckthorn seed meal. Seabuckthorn powder with 80 mesh standard drug sieve was obtained. Seabuckthorn powder alcohol extraction, the best technology is as follows: ethanol concentration of 75%, solid-liquid ratio of 1: 6, extraction temperature of 50 ° C, extracted three times each 60 min. Then, the extract of seabuckthorn was extracted by alkaline extraction acid precipitation method and hydrogen peroxide decolorization method. The technological parameters were as follows: pH = 10, solid-liquid ratio 1:12, alkaline extraction temperature 50 ℃, alkaline extraction 3 times for 50 min, The ratio of 30% hydrogen peroxide to alkali extract was 1:20, the decolorization temperature was 45 ℃, the decolorization time was 60 min, and the pH value was 5. Results: After three steps of crushing, sieving, alcohol, alkali and decolorization, the protein of sea buckthorn was obtained. The protein purity was 84.18% and the yield of the product was 11.39%. Conclusion: The extraction process is better.
