目的通过半定量逆转录-聚合酶链反应技术(RT-PCR)对急性白血病(acute leukemia,AL)患者及健康人群Apollon和COX-2这2种mRNA表达水平进行检测,探讨两者在AL诊断中的临床意义及在AL发生中的作用关系。方法常规收集40例新诊断的AL患者及20例健康人骨髓液或外周血标本,提取细胞总RNA,采用RT-PCR技术,内参照物选择β-actin,半定量检测靶mRNA的表达水平。结果新诊断的AL患者组Apollon mRNA表达水平为0.332±0.296,高于正常对照组表达水平0.026±0.018,差异有统计学意义(P<0.05);COX-2mRNA表达水平为0.916±0.154,高于正常对照组表达水平0.554±0.131,差异有统计学意义(P<0.05);两者的表达呈正相关(r=0.406,P<0.05)。结论 Apollon、COX-2mRNA在新诊断的AL患者中异常过度表达,并且呈正相关关系,两者的表达水平及相互作用可能是导致AL发生、发展的重要因素。
Objective To detect the mRNA expression levels of Apollon and COX-2 in patients with acute leukemia (AL) and healthy people by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) In the clinical significance and the role of AL in the relationship. METHODS: Forty freshly diagnosed AL patients and 20 healthy human bone marrow fluid or peripheral blood samples were collected routinely to extract the total RNA of the cells. RT-PCR and β-actin were selected as internal reference to detect the expression of target mRNA. Results The expression of Apollon mRNA in newly diagnosed AL patients was 0.332 ± 0.296, which was significantly higher than that in normal controls (0.026 ± 0.018, P <0.05). The expression level of COX-2 mRNA in AL patients was 0.916 ± 0.154, The expression level in normal control group was 0.554 ± 0.131, the difference was statistically significant (P <0.05). There was a positive correlation between the two expression levels (r = 0.406, P <0.05). Conclusions Apollon and COX-2 mRNA are abnormally overexpressed and positively correlated in newly diagnosed AL patients. The expression level and interaction of Apollon and COX-2 may be the important factors leading to the occurrence and development of AL.
