目的研究免疫性血小板减少症(ITP)患者外周血单核细胞(PBMC)中micro RNA-146a(mi R-146a)的表达水平及参芪益气生血合剂体外干预对其表达的影响。方法以荧光定量PCR法,对比ITP组与正常对照组PBMC中mi R-146a表达水平的差异,对比ITP药物干预组(参芪益气生血合剂)患者与ITP空白对照组患者PBMC中mi R-146a表达水平的差异。结果与正常对照组相比,ITP组患者PBMC中mi R-146a表达水平显著降低,差异具有统计学意义(P<0.05);ITP药物干预组患者PBMC中mi R-146a表达水平较ITP空白对照组高,且差异具有统计学意义(P<0.05)。结论 mi R-146a表达异常可能与ITP的发生发展相关,参芪益气生血合剂可能通过参与调节ITP患者的mi R-146a表达达到改善ITP效果。
Objective To study the expression of micro RNA-146a (mi R-146a) in peripheral blood mononuclear cells (PBMC) from patients with idiopathic thrombocytopenia (ITP) and the effect of Shenqi Yiqi Shengxue Mixture on its expression in vitro. Methods Compared with the expression of mi R-146a in PBMC of ITP group and normal control group by real-time quantitative PCR, we compared the expression of mi R-146a in PBMC of patients with ITP drug-intervention group and ITP blank control group, 146a expression level differences. Results Compared with the normal control group, the expression of mi R-146a in PBMC of ITP group was significantly lower than that in control group (P <0.05). The expression of mi R-146a in PBMC from ITP group was significantly lower than that of ITP control group Group height, and the difference was statistically significant (P <0.05). Conclusion The abnormal expression of mi R-146a may be related to the occurrence and development of ITP. Shenqi Yiqi Shengxue Mixture may improve the ITP effect by participating in the regulation of mi R-146a expression in ITP patients.
