目的研究环氧合酶-2抑制剂SC236对胃癌转移的影响,并初步探讨其作用机制。方法用人胃癌细胞株SGC7901建立裸鼠人胃癌原位移植瘤模型,随机分为实验组和对照组,分别经尾静脉注射SC236和生理盐水,观察各组生存期和肿瘤转移情况,并通过免疫组化方法检测移植瘤MVD、VEGF及MMP-2蛋白的表达。结果与对照组相比,SC236可明显延长荷瘤鼠生存期,抑制胃癌转移,降低移植瘤MVD,减低移植瘤VEGF及MMP-2蛋白的表达。结论SC236能有效抑制胃癌的转移,其机制可能与其抑制肿瘤的血管生成、减低MMP-2的表达有关。
Objective To study the effect of cyclooxygenase-2 inhibitor SC236 on gastric cancer metastasis and to investigate its mechanism. Methods Human gastric cancer orthotopically implanted tumor model was established with human gastric cancer cell line SGC7901 and randomly divided into experimental group and control group. SC236 and saline were injected through caudal vein to observe the survival and tumor metastasis of each group. Methods The expressions of MVD, VEGF and MMP-2 in tumor tissue were detected. Results Compared with the control group, SC236 could significantly prolong the survival of tumor-bearing mice, inhibit the metastasis of gastric cancer, reduce the MVD and reduce the expression of VEGF and MMP-2 in tumor xenografts. Conclusion SC236 can effectively inhibit the metastasis of gastric cancer, the mechanism may be related to its inhibition of tumor angiogenesis and reduce the expression of MMP-2.
