目的 研究 17β 雌二醇 (17β estradiol,E2 )对HOSTE85细胞骨形成的长时程效应。方法 3H 胸腺嘧啶参入法 (3H TdR)测细胞增殖 ;4 5Ca沉积法测细胞钙摄取 ;茜素红染色法测间质矿化。结果 E2 (0 1~ 10nmol·L- 1 )作用14d剂量依赖地刺激细胞3H TdR参入、增加4 5Ca摄取并增加茜素红染色面积。ICI1 82 ,780 (1 0nmol·L- 1 )能部分抑制E2作用 ,使其3H TdR参入分别减少了 36 5 6 % ,19 6 %和 15 4 % ,4 5Ca的摄入则分别减少了 2 2 2 7% ,37 2 8%和 4 0 1% ,茜素红染色面积减少。结论 E2 通过增加细胞数量、增加钙摄入而促进骨间质生成和矿化
Objective To study the long-term effects of 17β estradiol (E2) on the bone formation of HOSTE85 cells. Methods 3H thymidine incorporation assay (3H TdR) was used to measure the cell proliferation. Calcium uptake was measured by 4 5Ca deposition method. Alizarin red staining was used to determine the mineralization of interstitial cells. Results E2 (0 1 ~ 10nmol·L-1) stimulated the 3H TdR incorporation in a dose-dependent manner for 14 days, increased the uptake of 4Ca and increased the area of alizarin red staining. ICI1 82 and 780 (10 nmol·L-1) could partially inhibit the effect of E2, reducing their 3H TdR incorporation by 36 5 6%, 19 6% and 15 4%, respectively, while reducing 4 5 Ca up by 2 2 2 7%, 37 2 8% and 4 0 1%, alizarin red staining area decreased. Conclusions E2 promotes bone formation and mineralization by increasing cell numbers and increasing calcium intake
