1989年Tepper等把白细胞介素4(IL-4)基因转入乳腺癌细胞发现可以诱导机体特异性抗肿瘤作用,从而拉开了基因瘤苗研究的序幕.白细胞介素2(IL-2)是抗原活化的TH细胞分泌的细胞因子,在兔疫功能调节,尤其是在抗肿瘤免疫调节中起着重要的作用.近年来,国内外学者对IL-2基因瘤苗的研制作了大量的工作,本文就此进展作一综述.1 基因转移技术把IL-2基因转染肿瘤细胞,并获得较高水平的表达,是转基因瘤苗研究的关键技术.理想的技术方法能够使目的基因高效地导入人肿瘤细胞,并且操作简便,具有安全性.1.1 理化方法 包括电穿孔,颗粒轰击技术,DNA直接注射,脂质载体法,磷酸钙沉淀法,DETE-葡聚糖法等。磷酸钙沉淀法尚能形成稳定的整合和长期高效表达,在一定条件下仍不失为一个较为简便和有效的方法.脂质载体法是在体内外均可使用的方法.往黑色素瘤瘤体内直接注射脂质体包裹的IL-2基因10天
