目的 :定量地检测E 钙粘附蛋白 (E cad)及α 连接素 (α CTN)在膀胱移行细胞癌的表达 ,并探讨它们与肿瘤生物学行为的关系。方法 :应用流式细胞术 ,检测 30例膀胱癌冰冻组织的E cad和α CTN的表达量。结果 :肿瘤细胞E cad(0 .5 6 77± 0 .3194) ,α CTN(0 .5 72 6± 0 .2 895 )的表达量 (荧光指数 ,FI)均明显低于正常粘膜(P <0 .0 1)。浸润型肿瘤二者表达量低于表浅型肿瘤 (P <0 .0 1) ,复发者E cad表达量低于初发者 (P <0 .0 5 )。E cad和α CTN表达量呈正相关 (P <0 .0 1)。结论 :膀胱移行细胞癌E cad和α CTN表达均下调 ,并以浸润型肿瘤为显著 ,肿瘤复发后 ,其E cad表达量降低。E cad和α CTN表达量呈正相关。二者下调可能具有因果关系。
OBJECTIVE: To quantitatively detect the expression of E cadherin and α-CTN in transitional cell carcinoma of the bladder and to explore their relationship with the biological behavior of tumor. Methods: Flow cytometry was used to detect the expression of E cad and α CTN in 30 cases of bladder cancer frozen tissue. Results: The expression of E cad (0. 567 ± 0. 3194) and α CTN (0. 526 ± 0. 2895) in tumor cells were significantly lower than those in normal mucosa (P < 0 .0 1). The expression of both E-cadherin and E-cadherin in infiltrating tumors was lower than that in superficial tumors (P <0.01). The expression of E cad was lower in infiltrating tumors than in those without infiltrating tumors (P <0.05). E cad and α CTN expression was positively correlated (P <0.01). Conclusion: The expressions of E cad and α CTN were both down-regulated in transitional cell carcinoma of the bladder and the infiltrative tumors were significant. The expression of E cad decreased after tumor recurrence. E cad and α CTN expression was positively correlated. Both may have a causal relationship.
