目的:观察颗粒蛋白(progranulin,PGRN)阳性细胞在新生大鼠大脑皮层内的分布和PGRN在新生鼠大脑皮层神经元、星形胶质细胞和少突胶质细胞中的表达情况,探讨PGRN在新生鼠早期神经发育中的作用。方法:制备新生SD大鼠(1、7 d)脑组织切片,采用PGRN多克隆抗体进行免疫荧光组织化学染色法检测PGRN在新生鼠脑内的表达部位,免疫荧光组织化学双标法检测PGRN在新生鼠皮层内不同细胞中的定位。结果:新生鼠大脑皮质、侧脑室周围、胼胝体及海马中均表达PGRN。PGRN主要在新生鼠皮层内神经元中表达,而在星形胶质细胞和少突胶质细胞中几乎不表达。结论:新生鼠脑内表达PGRN,在大脑皮层内不同细胞表达模式有所不同,提示PGRN可能参与大鼠神经发育早期过程。
OBJECTIVE: To observe the distribution of progranulin (PGRN) positive cells in the cerebral cortex of newborn rats and the expression of PGRN in neurons, astrocytes and oligodendrocytes of neonatal rats, and to explore the role of PGRN in Role of Neonatal Rat in Early Neurodevelopment. Methods: The brain tissue sections of neonatal SD rats (1,7 d) were prepared. The expression of PGRN in the brain of neonatal rats was detected by immunofluorescence staining using PGRN polyclonal antibody. The immunofluorescence histochemical double-labeled method was used to detect the expression of PGRN Positioning in different cells in neonatal rat cortex. Results: PGRN was expressed in cortex, periventricular, corpus callosum and hippocampus of newborn rats. PGRN is mainly expressed in neonatal rat cortical neurons but scarcely expressed in astrocytes and oligodendrocytes. CONCLUSION: The expression of PGRN in the brain of neonatal rats is different in the expression pattern of different cells in the cerebral cortex, suggesting that PGRN may be involved in the early process of neural development in rats.
