本文用四唑盐(MTT)比色法和酶联免疫(ELISA)法研究了灵芝酮三醇对雄激素依赖性前列腺癌细胞株LNCaP细胞增殖和分泌前列腺特异性抗原(PSA)的抑制作用,另外还运用高效液相色谱法研究了灵芝酮三醇对5α-还原酶的抑制作用。结果显示,灵芝酮三醇对LNCaP细胞的增殖和分泌PSA的作用均呈现浓度依赖性抑制。另外,灵芝酮三醇对5α-还原酶的抑制作用也呈现浓度依赖性,作用浓度为2.5 mM时,其对5α-还原酶的抑制率为51.88%。结果表明灵芝酮三醇对LNCaP的抑制用既与其细胞毒性有关也与其对雄激素的抑制作用有关。
The inhibitory effect of ganoderma lucidum triol on the proliferation and secretion of prostate specific antigen (PSA) in androgen-dependent prostate cancer cell line LNCaP was studied by MTT assay and enzyme-linked immunosorbent assay (ELISA) In addition, the inhibitory effect of ganodermatriol on 5α-reductase was also studied by high performance liquid chromatography. The results showed that, Ganoderma lucidum ketone triol on LNCaP cells proliferation and secretion of PSA showed a concentration-dependent inhibition. In addition, ganodermatriol also showed a concentration-dependent inhibitory effect on 5α-reductase. When the concentration was 2.5 mM, the inhibitory rate of 5α-reductase was 51.88%. The results showed that the inhibitory effect of ganodermaketone on LNCaP was related to both its cytotoxicity and its inhibitory effect on androgen.
