目的:采用超高效液相色谱-电喷雾飞行时间串联质谱法(UHPLC-ESI-Q-TOF-MS/MS)对紫花地丁水提取物的主要化学成分进行分析鉴定。方法:采用Welch C18色谱柱(2.1 mm×100 mm,1.7μm),以0.1%甲酸水(A)-乙腈(B)为流动相梯度洗脱,流速0.35 m L·min~(-1),质谱采用电喷雾(ESI)离子源,在正负离子模式下采集数据,运行时间为30 min。结果:通过二级高分辨质谱分析结合对照品数据及相关文献,共鉴定32个化合物,包含香豆素、黄酮及有机酸类成分。结论:该研究利用UHPLC的高效分离能力和MS的高灵敏度检测优势,建立的分析方法,为控制紫花地丁药材质量、临床疗效及阐释其作用机制提供了科学依据。
OBJECTIVE: To identify and identify the main chemical constituents of aqueous extract from Viola yedoensi by ultra performance liquid chromatography-electrospray ionization time-of-flight mass spectrometry (UHPLC-ESI-Q-TOF-MS / MS). Methods: The mobile phase was eluted with a gradient of 0.1% formic acid (A) - acetonitrile (B) on a Welch C18 column (2.1 mm × 100 mm, 1.7 μm) with a flow rate of 0.35 m L · min -1 Mass spectrometry Electrospray (ESI) ion source was used to collect data in positive and negative ion mode and the running time was 30 min. RESULTS: Thirty-two compounds, including coumarin, flavonoids and organic acids, were identified by secondary high resolution mass spectrometry combined with reference data and related literature. Conclusion: This study utilized the advantages of UHPLC high-efficiency separation and MS high-sensitivity detection. The established analytical methods provided a scientific basis for controlling the quality of medicinal materials of Viola yedoensis, clinical efficacy and elucidating its mechanism of action.
