目的:探索以中药草乌抗肝癌有效成分———醚溶性生物碱为原料制备肝靶向白蛋白微球的可行性。方法:以常温乳化化学交联法制备微球,电子扫描显微摄影进行形态研究,酸性染料比色法测定包封率,动态透析法考察体外释药特性。结果:微球平均粒径0 .153μm ,内载药量为8 .6 ~9 .9μg·mg - 1 ,体外释放特性符合双指数双相动力学模型和单指数加Higuchi 平方根定律;室温条件下贮存3 个月质量稳定。结论:以中药草乌抗肝癌有效部位为原料制备白蛋白微球具有一定的可行性
OBJECTIVE: To explore the feasibility of preparing liver-targeting albumin microspheres using Chinese herbal medicine WuHuang active ingredients against liver cancer --- ether-soluble alkaloids as raw materials. Methods: The microspheres were prepared by emulsification-chemical crosslinking at room temperature. The morphology of the microspheres was observed by scanning electron microscopy. The entrapment efficiency was determined by acid dye colorimetric assay. The in vitro release characteristics were investigated by dynamic dialysis. Results: The average diameter of microspheres was 0.153μm, the internal drug loading was 8.6-6.9μg · mg - 1, and the in vitro release characteristics were in accordance with the double exponential biphasic kinetic model and single exponential plus Higuchi square root law. At room temperature Stored for 3 months and stable quality. Conclusion: It is feasible to prepare albumin microspheres by using the active fractions of Chinese herbal medicine “Wuwu” against liver cancer as raw materials
