背景与目的:研究表明,加热可提高兔VX2细胞对阿霉素(adriamycin,ADM)的增敏作用,可促进ADM进入细胞内,从而提高ADM的抗癌作用。作者在兔VX2细胞行ADM热化疗研究和兔肝VX2细胞移植瘤模型建立基础上,进行ADM兔肝动脉热灌注,以评价介入性化疗与介入性热化疗对兔肝癌的抑瘤效果。方法:将VX2细胞接种于60只新西兰白兔肝右叶,建立兔肝VX2移植癌模型。随机分4组,每组15只。利用导管经肝动脉分别给各组37℃生理盐水(第1组)、37℃ADM溶液(第2组)、60℃生理盐水(第3组)、60℃ADM溶液(第4组),1周后观察各组肿瘤体积及血清AST水平,观察荷瘤兔的存活期。结果:第4组肿瘤生长率(0.53±0.21)%,与第1组(3.48±1.17)%相比有显著性差异(P<0.01),与第2组的(1.09±0.26)%、第3组的(3.32±1.28)%相比亦有显著性差异(P<0.05)。第4组存活期(87.0±2.0)天,与第1组(40.5±3.0)天相比差异有显著性(P<0.05)。第4组血清AST水平给药前后变化与其它各组相比无显著性差异(P>0.05),与对第1组相比有显著性差异(P<0.05)。结论:60℃ADM溶液热灌注可降低肿瘤生长率,延长存活期,而对肝功能的损害是可逆的。
BACKGROUND AND PURPOSE: The study shows that heating can enhance the sensitization of rabbit VX2 cells to adriamycin (ADM), promote the entry of ADM into cells and increase the anticancer effect of ADM. On the basis of ADM thermochemotherapy and rabbit liver VX2 cell xenograft model in rabbit VX2 cells, we performed thermal perfusion of hepatic artery in ADM rabbit to evaluate the anti-tumor effect of interventional chemotherapy and interventional thermochemotherapy on rabbit liver cancer. Methods: VX2 cells were inoculated into the right lobe of 60 New Zealand white rabbits to establish a rabbit VX2 transplanted carcinoma model. Randomly divided into 4 groups, each group of 15. The rats in each group were treated with 37 ℃ normal saline (group 1), 37 ℃ ADM solution (group 2), 60 ℃ saline (group 3), 60 ℃ ADM solution (group 4), 1 Week after the observation of tumor volume and serum AST levels were observed in the survival of tumor-bearing rabbits. Results: The tumor growth rate of group 4 was significantly higher than that of group 1 (3.48 ± 1.17)% (0.53 ± 0.21)% and (1.09 ± 0.26)%, respectively There was also a significant difference between the three groups (3.32 ± 1.28)% (P <0.05). There was a significant difference between the first group (40.5 ± 3.0) days and the fourth group (87.0 ± 2.0) days (P <0.05). There was no significant difference between the 4th group before and after administration of AST (P> 0.05) and the first group (P <0.05). CONCLUSION: Thermal perfusion with 60 ° C ADM solution reduces tumor growth and prolongs survival, whereas damage to liver function is reversible.
