目的研究姜油和6-姜酚对小鼠肝脏UGT、SULT1A1和GST活性及其mRNA表达的影响。方法给小鼠灌服不同剂量的姜油和溶媒对照液,每天2次,连续3天。以分光光度法,检测肝脏中上述代谢酶活性;以RT-PCR法,检测UGT1A1/SULT1A1mRNA在肝脏中的表达。结果姜油(15 mg·kg-1)对小鼠UGT和GST活性有显著的诱导作用(P<0.05或P<0.01);以2-萘酚和4-硝基酚为底物测定SULT1A1活性时,姜油(15,3 mg·kg-1)对SULT1A1均有明显的诱导作用(P<0.05或P<0.01);RT-PCR结果显示,仅有10 mg·kg-1的6-姜酚能够提高UGT1A1/SULT1A1 mRNA的表达。结论临床用药期间,应注意食用姜类产品对Ⅱ相药物代谢酶的影响。
Objective To study the effects of Ginger oil and 6-gingerol on the activity of UGT, SULT1A1 and GST in mouse liver and their mRNA expression. Methods Mice were fed different doses of ginger oil and vehicle control solution twice daily for 3 consecutive days. The above metabolic enzymes activity in liver was detected by spectrophotometry. The expression of UGT1A1 / SULT1A1 mRNA in liver was detected by RT-PCR. Results Ginger oil (15 mg · kg -1) had a significant effect on the activity of UGT and GST in mice (P <0.05 or P <0.01), and SULT1A1 activity was measured by 2-naphthol and 4-nitrophenol (15, 3 mg · kg-1) induced SULT1A1 significantly (P <0.05 or P <0.01). The results of RT-PCR showed that only 10 mg · kg-1 of 6-ginger Phenol can increase UGT1A1 / SULT1A1 mRNA expression. Conclusion During clinical use, attention should be paid to the influence of ginger products on the enzyme metabolism of phase Ⅱ drugs.
