目的:建立痛痹康胶囊中士的宁的含量测定方法,为其质量控制提供参考依据。方法:采用高效液相法,色谱柱采用正相硅胶柱(4.6mm×250mm,5μm),以正己烷-二氯甲烷-无水甲醇-三乙胺(47.5∶47.5∶5∶0.4)为流动相,流速为1.0mL/min,检测波长为280nm,进样量为10μL。结果:士的宁的回归方程为y=7.108 34x-21.971 91,r=0.999 86,其在31.00~387.40μg/mL浓度范围内与峰面积积分线性关系良好,平均加样回收率为101.8%(n=6),RSD值为2.15%;样品中士的宁的平均含量为0.19mg/粒。结论:该方法操作简单,稳定性、重现性好,可用于该制剂中士的宁含量的检测。
Objective: To establish a method for determining the content of strychnine in Tongbikang capsule, and to provide a reference for its quality control. Methods: The mobile phase consisted of n-hexane-dichloromethane-absolute methanol-triethylamine (47.5:47.5:5:0.4), using a normal phase silica gel column (4.6 mm × 250 mm, 5 μm) Phase, flow rate 1.0mL / min, detection wavelength 280nm, injection volume 10μL. Results: The regression equation of strychnine was y = 7.108 34x-21.971 91, r = 0.999 86, which showed a good linear relationship with the integral of peak area in the range of 31.00-387.40μg / mL with an average recovery of 101.8% n = 6) with a RSD value of 2.15%. The average content of strychnine in the sample was 0.19 mg / tablet. Conclusion: The method is simple, stable and reproducible. It can be used for the determination of strychnine in the preparation.
