目的建立同时测定大鼠血浆中茶碱、甲硝唑的HPLC-UV分析方法,探索茶碱与甲硝唑在大鼠体内是否存在药动学上的相互作用。方法将18只雄性SD大鼠随机分成氨茶碱组、甲硝唑组以及氨茶碱和甲硝唑联合给药组,于给药后不同时间点采集血样,HPLC测定二者的血药浓度,采用DAS2.0软件计算药动学参数,并用SPSS19.0软件对其进行单因素方差分析,以95%的可信区间判断药物是否发生相互作用。结果氨茶碱和甲硝唑联合用药组与单用药组药动学参数t1/2,MRT,AUC0-t,Vd和CL均无显著性差异(P>0.05)。结论氨茶碱和甲硝唑联合用药后不产生明显的药动学相互作用。
OBJECTIVE To establish a HPLC-UV method for the simultaneous determination of theophylline and metronidazole in rat plasma and to explore the pharmacokinetic interaction between theophylline and metronidazole in rats. Methods Eighteen male Sprague-Dawley rats were randomly divided into aminophylline group, metronidazole group, aminophylline group and metronidazole group. Blood samples were collected at different time points after administration, and plasma concentrations of both drugs The pharmacokinetic parameters were calculated by DAS2.0 software. One-way analysis of variance (ANOVA) was performed using SPSS19.0 software to determine whether the drug interacted with 95% confidence interval. Results There was no significant difference in pharmacokinetic parameters t1 / 2, MRT, AUC0-t, Vd and CL between aminophylline and metronidazole combination group and single drug group (P> 0.05). Conclusion The combination of aminophylline and metronidazole did not produce significant pharmacokinetic interaction.
