目的:小鼠睾丸生殖细胞瘤易感蛋白DND1是在进化中保守的RNA结合蛋白。为了进一步了解DND1的功能,我们筛选了与DND1相互作用的蛋白。方法:采用酵母双杂交系统,用Dnd1基因编码序列构建诱饵载体筛选小鼠10.5天的胚胎文库。结果:筛选小鼠胚胎cDNA文库得到4个与DND1相互作用蛋白,分别为Jun蛋白、辅肌动蛋白α、核受体结合蛋白和氯离子通道2。结论:DND1可能参与调节细胞増殖、迁移及细胞转化等过程。
Objective: Mouse testicular germ cell tumor susceptibility protein DND1 is an evolutionarily conserved RNA-binding protein. To further understand the function of DND1, we screened for proteins that interact with DND1. Methods: The yeast two-hybrid system was used to construct the bait vector with Dnd1 gene coding sequence to screen the mouse embryonic library for 10.5 days. RESULTS: Four DND1 interacting proteins were screened from mouse embryonic cDNA library, which were Jun protein, actin α, nuclear receptor binding protein and chloride channel 2. Conclusion: DND1 may be involved in the regulation of cell proliferation, migration and cell transformation.
