[目的]建立啤酒中氨基酸的高效阴离子交换色谱分离-脉冲安培检测法测定。[方法]样品用去离子水稀释后经滤膜过滤直接进样,采用梯度洗脱、阴离子交换色谱-积分脉冲安培检测法测定啤酒中的氨基酸。[结果]5种品牌的啤酒氨基酸平均含量为773.54mg/L,其中氨基酸含量最高的为1040.72mg/L,最低的为532.77mg/L。[结论]不同原料、不同酿造方式使啤酒中的氨基酸含量各不相同;利用阴离子交换色谱分离-脉冲安培检测法测定啤酒中的氨基酸,样品前处理简单,方法适用性宽,测定重复性好,灵敏度高,定性定量准确,结果满意。
[Objective] The research aimed to establish an efficient anion exchange chromatography of amino acids in beer - pulsed amperometric detection. [Method] The samples were diluted with deionized water, filtered through a membrane filter, and then directly injected into the sample. Gradient elution and anion - exchange chromatography - integrated pulsed amperometric detection were used to determine the amino acids in beer. [Result] The average content of amino acids in the five brands of beer was 773.54 mg / L, among which the highest content of amino acids was 1040.72 mg / L and the lowest was 532.77 mg / L. [Conclusion] The contents of amino acids in beer varied with different raw materials and brewing methods. The amino acids in beer were determined by anion exchange chromatography - pulse amperometric detection. The sample preparation was simple, the method was suitable for wide range of applications, High sensitivity, qualitative and quantitative accurate, the results are satisfactory.
