目的:研究脉君安片对自发性高血压大鼠相关组织中内皮素-1(ET-1)和一氧化氮合酶(NOS)表达的影响,探讨其降压机制。方法:12只成年自发性高血压大鼠随机分为给药组和对照组,给药组灌胃模拟脉君安片(氢氯噻嗪10mg·kg-1.d-1+葛根素36mg·kg-1.d-1+钩藤总碱122mg·kg-1.d-1),对照组灌胃等量去离子水。每周测血压1次,4周后处死动物,分别检测组织与血浆中ET-1和NOS mRNA和蛋白质的表达量。结果:与对照组相比,用药4周后给药组降压效果显著(P<0.05),肝、肾与血浆中NOS基因与蛋白表达水平上调,而ET-1的基因与蛋白表达水平下调。结论:脉君安片可通过上调NOS的表达和下调ET-1的表达,降低自发性高血压大鼠的血压。
OBJECTIVE: To study the effect of Maijunan Tablet on the expression of endothelin-1 (ET-1) and nitric oxide synthase (NOS) in the related tissues of spontaneously hypertensive rats and to explore its antihypertensive mechanism. METHODS: Twelve adult spontaneously hypertensive rats were randomly divided into two groups: the administration group and the control group. The injection group was given intragastrically simulated Qiujunan tablets (hydrochlorothiazide 10 mg·kg-1.d-1+puerarin 36 mg·kg-1. .d-1+ rhynchophylline 122 mg·kg-1.d-1). The control group was given equal volume of deionized water by gavage. Blood pressure was measured once a week, and animals were sacrificed 4 weeks later. Tissue and plasma ET-1 and NOS mRNA and protein expression levels were measured. Results: Compared with the control group, the antihypertensive effect was significant (P<0.05) after 4 weeks of treatment. The expression of NOS gene and protein in liver, kidney and plasma was up-regulated, while the expression of ET-1 was down-regulated. . Conclusion: Maijunan Tablets can reduce the blood pressure of spontaneously hypertensive rats by up-regulating the expression of NOS and down-regulating the expression of ET-1.
