考察了所制备的带组氨酸配基的尼龙66亲和膜在不同离子强度、pH值的盐溶液中及不同温度和流速下对内毒素的去除效果,并对这种亲和介质与内毒素的相互作用机理进行了研究。结果表明:在盐溶液离子强底低于02mol/L、pH值3~8的范围及温度低于50℃、流速小于5mL/min下,可获得最佳去除效果。在选择的最佳条件下能有效去除牛血清白蛋白、氨基酸注射液、人血样等中的内毒素。去除率大部分在90%以上,并能保留住这些样品溶液中绝大多数有效组分,回收率一般都高于80%。
The effect of removing histidine-based Nylon 66 affinity membrane on the removal of endotoxin in salt solution with different ionic strength and pH value and at different temperatures and flow rates was investigated. The interaction mechanism of toxins has been studied. The results showed that the optimum removal efficiency was obtained when the salt concentration was less than 0.2 mol / L, the pH range was 3 ~ 8, the temperature was lower than 50 ℃ and the flow rate was less than 5 mL / min. In the best choice of the conditions can effectively remove endotoxin in bovine serum albumin, amino acid injection, human blood samples. Most of the removal rate is above 90%, and can retain most of the active components in these sample solutions, the recovery rates are generally higher than 80%.
