目的观察雷公藤红素对ATRA诱导白血病细胞分化成熟的影响。方法用细胞形态学、生化指标、分子标志等指标,观察雷公藤红素和/或ATRA不同处理组NB4分化情况。结果与对照组相比,ATRA诱导后,NB4细胞呈现中性粒细胞分叶核形态;NBT还原能力明显增加;分化抗原CD13和CD33表达下降,而CD11b和CD15表达增加。雷公藤红素与ATRA联合应用,结果与ATRA处理组相似。结论雷公藤红素不影响ATRA诱导NB4细胞分化成熟作用,提示雷公藤红素作为分化综合征的治疗和预防药物值得进一步研究。
Objective To observe the effects of tripterine on differentiation and maturation of ATRA-induced leukemia cells. Methods The cell morphology, biochemical indicators, molecular markers and other indicators to observe the triptolide and / or ATRA different treatment group NB4 differentiation. Results Compared with the control group, NB4 cells showed neutrophilic lobular morphology after NBT cells were induced by ATRA. The NBT reduced ability was significantly increased. The expression of CD13 and CD33 decreased, while the expression of CD11b and CD15 increased. Tripterine combined with ATRA results similar to the ATRA treatment group. Conclusion Tripterine does not affect ATRA-induced NB4 cell differentiation and maturation, suggesting that tripterine as a therapeutic and prophylactic drug for differentiation syndrome deserves further study.
