目的:观察转化生长因子β1(TGF-β1)、骨形成蛋白-2(BMP-2)在人下颌骨骨折骨痂组织中的表达及量的变化。方法:在下颌骨骨折的手术患者中,收集术中去除的骨断端间骨痂组织30例,以2例方颌畸形患者术中切取的下颌角骨块作正常对照;利用SABC免疫组织化学方法检测骨折后不同时期骨痂组织及正常下颌角骨组织中TGF-β1、BMP-2的表达。结果:TGF-β1、BMP-2在正常骨组织中没有表达;在不同时期骨痂组织中均有表达,TGF-β1在骨折后第1周到第3周表达量缓慢增加,至第3周时达到高峰,随着时间推移,表达量逐渐下降;BMP-2在骨折后前2周表达呈增加趋势,第2周达到高峰,以后随时间推移,表达量缓慢下降。结论:①BMP-2可能是启动骨折修复的因子之一;②TGF-β1可能是骨折修复的另一条信号通路;③BMP-2与TGF-β1在骨折愈合过程中可能存在协同作用。
OBJECTIVE: To observe the expression of BMP-2 and transforming growth factor β1 (TGF-β1) and their quantity in callus tissue of human mandibular fracture. Methods: Thirty patients with mandibular fractures underwent surgical resection. Thirty patients with intramedullary callus were collected during surgery. Two cases of mandibular angle fractures were performed as normal controls. The SABC immunohistochemistry Methods The expressions of TGF-β1 and BMP-2 in callus tissue and normal mandibular angle tissue at different stages after fracture were detected. Results: TGF-β1 and BMP-2 were not expressed in normal bone tissue. The expression of TGF-β1 and BMP-2 was expressed in callus tissue at different time points. The expression of TGF-β1 increased slowly from the first week to the third week after fracture. Reached a peak, with the passage of time, the expression gradually decreased; BMP-2 expression increased in the first 2 weeks after fracture, peaked in the 2nd week, and then decreased slowly over time. Conclusion: ①BMP-2 may be one of the factors to initiate fracture repair; ②TGF-β1 may be another signal pathway for fracture repair; ③BMP-2 and TGF-β1 may have synergistic effect during fracture healing.
