为了确立PCR单管法快速检测单纯疱疹病毒(HSV-1)的临床价值,本文对18例经临床、EEG,MRI或CT诊断的单疱病毒脑炎(HSE)患者24份CSF标本,先经解偶联裂解吸附预处理,抽提模板DNA,然后进行PCR一次性扩增研究。首次扩增18份CSF标本。其中17份PCR阳性,1份阴性,但6天后复查转为阳性。5例PCR阳性患儿经正规抗病毒治疗后均转为阴性。对照组30例其他中枢神经系统疾病均为阴性。结果表明,该PCR单管法具有灵敏、简便、快速、价廉和污染机会少等优点,特别适用于检测婴幼儿患者。
In order to establish the clinical value of HSV-1 assay by PCR single tube method, 24 CSF samples from 18 herpes simplex virus encephalitis (HSE) patients diagnosed by clinical, EEG, MRI or CT, Unconjugate cleavage adsorption pretreatment, extraction of template DNA, and then one-time amplification PCR study. 18 CSF samples were amplified for the first time. Of these, 17 were PCR-positive and one was negative, but returned to positive after 6 days. 5 cases of PCR-positive children after formal antiviral therapy turned negative. 30 cases of other central nervous system diseases in the control group were negative. The results showed that the PCR tube method is sensitive, simple, rapid, inexpensive and less chance of contamination, especially for the detection of infants and young children.
