目的:研究葡萄糖调节蛋白78(glucose-regulated protein 78,GRP78)在胆管癌细胞中的表达及其对胆管癌细胞增殖和迁移的影响。方法:体外培养胆管癌细胞QBC939、RBE和HCCC-9810,应用Western blot检测GRP78的表达,用CCK-8比色法和Transwell小室迁移实验检测GRP78对胆管癌细胞增殖和迁移的影响。结果:胆管癌细胞中GRP78呈高表达,且GRP78的高表达不依赖于内质网应激。干扰GRP78表达显著抑制胆管癌细胞的增殖和迁移(P<0.05)。结论:GRP78对胆管癌细胞的增殖和迁移起重要促进作用,GRP78在胆管癌治疗中具有潜在的靶点价值。
Objective: To investigate the expression of glucose-regulated protein 78 (GRP78) in cholangiocarcinoma cells and its effect on the proliferation and migration of cholangiocarcinoma cells. Methods: The cholangiocarcinoma cells QBC939, RBE and HCCC-9810 were cultured in vitro. The expression of GRP78 was detected by Western blot. The effect of GRP78 on the proliferation and migration of cholangiocarcinoma cells was detected by CCK-8 assay and Transwell migration assay. Results: GRP78 was highly expressed in cholangiocarcinoma cells, and the high expression of GRP78 did not depend on endoplasmic reticulum stress. Interference with GRP78 expression significantly inhibited the proliferation and migration of cholangiocarcinoma cells (P <0.05). CONCLUSION: GRP78 plays an important role in the proliferation and migration of cholangiocarcinoma cells. GRP78 has potential target value in the treatment of cholangiocarcinoma.
