利用国内15个代表性自交系及15个来自国家区域试验的玉米杂交种进行实验室复筛,并对玉米数据库MaizeGDB上已公布的1900个玉米SSR(simple sequence repeats)引物进行文献初筛,确定了500个高多态性引物,建立了玉米高多态性引物遗传图谱;并按照在玉米全基因组均匀分布的原则,从每条染色体上选取10个引物,累计100个,作为一套通用的SSR核心引物,推荐供今后一般研究优先选用.对这套核心引物按照统一的标准进行重新设计并构建基于毛细管五色荧光检测系统的高通量多重PCR复合扩增体系,已经建立了两套10重PCR组合,每套组合均由每条染色体上选取1个引物,累计10个构成.
A total of 1900 maize SSR primers published on the MaizeGDB database were preliminarily screened using 15 representative inbred lines from China and 15 maize hybrids from national regional trials. A total of 500 highly polymorphic primers were identified and a genetic map of high polymorphic primers was established. Ten primers were selected from each chromosome according to the principle of uniform genome-wide distribution in maize. Of SSR core primers recommended for future general research priority selection of the core primers in accordance with a unified standard redesign and construction based on capillary five-color fluorescence detection system of high-throughput multiplex PCR multiplex amplification system, has established two sets of 10 Heavy PCR combination, each combination of each chromosome by selecting a primer, a total of 10 components.
