目的 探讨变异型p53蛋白表达对人乳腺癌细胞阿霉素抗药性的影响。方法 应用琼脂培养MTT法检测阿霉素 (ADM)对 9例人乳腺癌新鲜标本药物敏感性。用S -P免疫组化技术观察变异型p53蛋白表达 ,并用ICM - 10 0细胞DNA图像分析系统作半定量分析 ;同时观察阿霉素对人乳腺癌手术前化疗疗效。结果 (1) 9例人乳腺癌新鲜标本中 7例变异型p53蛋白表达阳性 (阳性率为 77.77% )。 (2 )变异型p53蛋白表达量 (阳性面积占总面积百分比 )与人乳腺癌细胞阿霉素抗药指数 (人乳腺癌新鲜标本阿霉素的IC50值与 1/ 10阿霉素血浆峰值浓度的比值 )呈正相关 (r =0 .7956 ,P <0 .0 1)。 (3)变异型p53蛋白表达量与人乳腺癌患者手术前阿霉素化疗疗效积分呈负相关 (r =- 0 .86 37,P <0 .0 1)。结论 变异型p53蛋白表达与人乳腺癌细胞阿霉素抗药性相关
Objective To investigate the effect of variant p53 protein on the resistance of human breast cancer cells to adriamycin. Methods MTT assay was used to detect the sensitivity of adriamycin (ADM) to 9 samples of fresh human breast cancer. S-P immunohistochemistry technique was used to observe the expression of variant p53 protein and semi-quantitative analysis was performed using ICM-100 cell DNA image analysis system. At the same time, the efficacy of doxorubicin on preoperative breast cancer was observed. Results (1) The expression of p53 protein was positive in 7 cases of fresh human breast cancer (positive rate was 77.77%). (2) Expression of variant p53 protein (percentage of positive area in total area) and doxorubicin resistance index of human breast cancer cells (IC50 value of human breast cancer fresh specimen doxorubicin and 1/10 doxorubicin plasma peak concentration The ratio is positively correlated (r = 0.7956, P < 0.01). (3) The expression level of variant p53 protein was negatively correlated with the score of adriamycin chemotherapy before surgery in patients with breast cancer (r =- 0.867, P < 0.01). Conclusion The expression of variant p53 protein is related to the drug resistance of human breast cancer cell doxorubicin
