目的观察活脉通络饮对脑缺血再灌注大鼠脑组织Caspase-3表达的影响,探讨活脉通络饮对脑组织保护作用的机制。方法 Wistar大鼠随机分为正常对照组、缺血再灌注组、活脉通络饮低剂量组、活脉通络饮中剂量组、活脉通络饮高剂量组和尼莫地平阳性对照组。采用改良的线栓法,建立大鼠大脑中动脉栓塞(MCAO)脑缺血再灌注模型。采用HE染色观察大鼠脑组织形态改变,免疫组织化学法和Western Blot定性和定量法检测脑组织中Caspase-3的表达。结果缺血再灌注组大鼠脑组织Caspase-3的表达水平较正常对照组明显增加(<0.01),活脉通络饮组与缺血再灌注组比较Caspase-3蛋白表达明显减少(<0.01),而与尼莫地平阳性对照组相比没有明显差异。结论活脉通络饮的脑保护作用的机制之一可能与其下调Caspase-3表达相关。PP
Objective To observe the effects of Huomai Tongluo drink on the expression of Caspase-3 in brain tissue of rats with cerebral ischemia-reperfusion injury, and to explore the mechanism of Huo Mai Tongluo drink on brain tissue. Methods Wistar rats were randomly divided into normal control group, ischemia-reperfusion group, low-dose Huoxuetongluo decoction group, middle-dose Huoxuetongluo decoction group, high-dose Huoxuetongluo decoction group and nimodipine positive control group . A rat model of middle cerebral artery occlusion (MCAO) cerebral ischemia-reperfusion was established by modified suture method. The morphological changes of rat brain were observed by HE staining. The expression of Caspase-3 in brain tissue was detected by immunohistochemistry and Western Blot. Results The expression of Caspase-3 in brain tissue of ischemia-reperfusion group was significantly higher than that of normal control group (<0.01), Caspase-3 protein expression was significantly decreased in Huomai Tongluo drink group compared with ischemia reperfusion group (<0.01 ), But no significant difference compared with nimodipine positive control group. Conclusion One of the mechanisms of brain protective effect of Huatei Tongluo drink may be related to its downregulation of Caspase-3 expression. PP
