目的:测定复方根皮素乳膏中根皮素、藁本内酯的离体大鼠透皮吸收率。方法:以Franz智能透皮实验仪进行体外透皮吸收试验,取不同时间点接收液,以HPLC法测定根皮素、藁本内酯含量,计算累计释放量和累计透过量,进行透皮吸收行为评价。结果:根皮素、藁本内酯分别在0.044 2~4.42μg·ml~(-1)(r=0.999 9)和0.048 7~4.87μg·ml~(-1)(r=0.999 9)范围内线性良好,平均回收率分别为99.85%、99.92%,RSD分别为0.67%、0.66%(n=9),根皮素、藁本内酯透过离体大鼠皮肤的皮肤渗透行为符合Higuchi方程,r值均大于0.9,即累积渗透量Qn与时间t呈良好线性关系。结论:复方根皮素乳膏具有良好的透皮性能。
Objective: To determine the transdermal absorptivity of isolated rat phlebitis and ligustilide from compound phloretin. Methods: Percutaneous transdermal absorption test was performed with Franz intelligent transdermal test apparatus. The liquid samples were collected at different time points. The contents of phloretin and ligustilide were determined by HPLC. The cumulative release and cumulative permeation were calculated and transdermally absorbed Behavior evaluation. Results: The contents of phloretin and ligustilide in the range of 0.044 2 ~ 4.42 μg · ml ~ (-1) (r = 0.999 9) and 0.048 7 ~ 4.87 μg · ml ~ (-1) (r = 0.999 9) The average recoveries were 99.85% and 99.92%, respectively, with RSDs of 0.67% and 0.66%, respectively (n = 9). Skin permeation of phloretin and ligustilide through isolated rat skin was in line with Higuchi’s Equation, r values ?? are greater than 0.9, that is, cumulative infiltration Qn and time t showed a good linear relationship. Conclusion: The compound phloretin cream has good transdermal properties.
