目的探讨CCND1、MDM2在乳腺癌中的表达特点及其表达与预后的关系。方法采用免疫组织化学SP法检测CCND1、MDM2在58例乳腺癌及19例正常乳腺组织中的表达,分析两者的表达情况与疾病的临床病理特征之间的关系及蛋白间的相关性,并分析58例乳腺癌患者的随访资料,判断CCND1、MDM2的表达与患者淋巴结转移情况及预后的关系。结果在乳腺癌和正常乳腺组织中,CCND1蛋白的阳性表达率分别为56.9%(33/58)、15.8%(3/19);MDM2蛋白的阳性表达率分别为81.0%(47/58)、21.1%(4/19)。两组中CCND1和MDM2蛋白的阳性表达率均有统计学意义(P<0.05)。CCND1和MDM2蛋白的表达与乳腺癌腋窝淋巴结转移相关(P<0.05),与年龄、肿瘤大小、TNM分期无关(P>0.05)。在淋巴结转移病例中,CCND1蛋白表达与生存期显著相关(P<0.05);在无淋巴结转移的病例中,MDM2蛋白的表达与生存期显著相关(P<0.05)。CCND1蛋白表达与MDM2蛋白的表达相关且具有一致性(χ2=17.918,P<0.05,Kappa=0.472)。结论 CCND1与MDM2蛋白在乳腺癌组织中的异常表达,说明其可能共同参与了乳腺癌的发生、发展和转移,并与患者预后密切相关。
Objective To investigate the expression of CCND1 and MDM2 in breast cancer and its relationship with prognosis. Methods The immunohistochemical SP method was used to detect the expression of CCND1 and MDM2 in 58 cases of breast cancer and 19 cases of normal breast tissue. The relationship between the expression of CCND1 and MDM2 and the clinicopathological features of the disease and the correlation between the proteins were analyzed. The follow-up data of 58 cases of breast cancer were analyzed to determine the relationship between the expression of CCND1 and MDM2 and lymph node metastasis and prognosis. Results The positive rates of CCND1 protein in breast cancer and normal breast tissue were 56.9% (33/58) and 15.8% (3/19), respectively. The positive rates of MDM2 protein were 81.0% (47/58) 21.1% (4/19). The positive rates of CCND1 and MDM2 in both groups were statistically significant (P <0.05). The expression of CCND1 and MDM2 protein were correlated with axillary lymph node metastasis in breast cancer (P <0.05), but not with age, tumor size and TNM stage (P> 0.05). In the cases of lymph node metastasis, the expression of CCND1 protein was significantly correlated with the survival time (P <0.05). In the cases without lymph node metastasis, the expression of MDM2 protein was significantly correlated with the survival time (P <0.05). CCND1 protein expression and MDM2 protein expression related and consistent (χ2 = 17.918, P <0.05, Kappa = 0.472). Conclusion The abnormal expression of CCND1 and MDM2 protein in breast cancer indicates that they may participate in the occurrence, development and metastasis of breast cancer, which is closely related to the prognosis of patients.
