扫描电镜观察细胞组织和器官的内部结构要采用断裂方法。冷冻断裂法、树脂断裂法等,是常用的方法,但这些方法有组织断裂面小,断裂部位不易控制,使用的药品和液氮来源受限,步骤较为繁杂等缺点。我们将Carrl氏冰冻切片法略加改进,应用新鲜组织冷冻切面制样进行扫描电镜观察,经过一年试用,发现此法有检查目标定位准确;可与冰冻切片HE染色组织学图象相对照;标本面积大;方法简单易行;扫描图象稳定等优点。现将此法总结如下:
Scanning electron microscopy to observe the internal structure of tissues and organs to adopt the fracture method. Freezing fracture method and resin fracture method are common methods.
