目的 通过对口腔正常组织与混合瘤、高分化粘液表皮样癌及颊癌中的M期促进因子(M phasepromotingfactor,MPF)的存在状态及活性的比较 ,探讨MPF与口腔肿瘤的发展及恶性度间的关系。方法 利用免疫印迹法及Gollicano法测定MPF的量并进行活性测定分析。结果 MPF的 2个亚基cdc2及CyclinB都存在于口腔正常组织、混合瘤、高分化粘液表皮样癌及颊癌中 ,且肿瘤组织中的MPF量高于正常组织 ;肿瘤组织中的MPF的活性量高于正常组织。颊癌较正常组织高 6 4% ,提示很可能MPF的活性变化与肿瘤的恶性度有关。结论 MPF存在于正常组织、混合瘤、高分化粘液表皮样癌及颊癌中 ,肿瘤组织中的量及活性均高于正常组织。蛋白激酶C可激活MPF ,在肿瘤组织中蛋白激酶C可能通过激活MPF来促进肿瘤的增殖。MPF的活性可能与肿瘤的发展及恶性度相关。
OBJECTIVE: To compare the existence and activity of M phase promoting factor (MF) in normal oral mucosa and mixed tumor, well differentiated mucoepidermoid carcinoma and buccal carcinoma, and to explore the relationship between the development of MPF and oral tumor and the relationship between malignant degree Relationship. Methods The amount of MPF was determined by immunoblotting and Gollicano method and the activity of MPF was assayed. Results The two subunits of MPF, cdc2 and CyclinB, were found in normal oral mucosa, mixed tumor, highly differentiated mucoepidermoid carcinoma and buccal carcinoma, and the MPF content in tumor tissue was higher than that in normal tissue. The activity of MPF in tumor tissue Higher than normal tissue. Cheek cancer was 64% higher than normal tissues, suggesting that MPF activity may be related to the degree of malignancy of the tumor. Conclusions MPF is present in normal tissues, mixed tumors, highly differentiated mucoepidermoid carcinomas and buccal carcinomas. The amount and activity of MPF in tumor tissues are higher than those in normal tissues. Protein kinase C activates MPF, and protein kinase C may promote tumor proliferation by activating MPF in tumor tissues. The activity of MPF may be related to tumor development and malignancy.
