目的探讨双氢青蒿素对Wistar大鼠放射性肺炎(RP)的治疗作用。方法 60只雄性Wistar大鼠随机分成3组:空白组、模型组、双氢青蒿素(DHA)组。用6MV-X线直线加速器对大鼠双肺进行单次照射15Gy,照射后从第1天开始灌胃给药,连续28d,分别于照射后7d,14d,21d,28d处死。测定各组大鼠血液及肺泡灌洗液中红细胞和白细胞总数;大鼠肺脏组织学观察;ELISA法测血清中NF-κB,TNF-α和IL-6的表达。结果空白组大鼠肺表面光滑、无炎症反应;DHA组大鼠肺组织较模型组炎症反应轻,模型组肺组织炎症反应最明显。DHA组肺泡灌洗液中白细胞计数及血清中NF-κB,TNF-α和IL-6的表达均较模型组低,差异有统计学意义(P<0.05)。结论 DHA可通过降低细胞因子NF-κB,TNF-α和IL-6的表达水平,减轻炎症反应,起到治疗放射性肺炎的作用。
Objective To investigate the therapeutic effect of dihydroartemisinin on radiation pneumonitis (RP) in Wistar rats. Methods Sixty male Wistar rats were randomly divided into 3 groups: blank group, model group and dihydroartemisinin (DHA) group. Rats were irradiated with a single dose of 15Gy for 6 weeks with a 6MV-X linear accelerator. The rats were administered intragastrically once a day for 28 days and were sacrificed at 7d, 14d, 21d and 28d after irradiation respectively. The total number of erythrocytes and leukocytes in the blood and bronchoalveolar lavage fluid of rats in each group was determined. The lung histology was observed. The expressions of NF-κB, TNF-α and IL-6 were detected by ELISA. Results The lungs of the rats in the blank group were smooth and had no inflammatory reaction. The lung tissue of the DHA group was lighter than the model group and the inflammatory reaction of the model group was the most obvious. The leukocyte counts and the expressions of NF-κB, TNF-α and IL-6 in the alveolar lavage fluid in DHA group were lower than those in the model group, with statistical significance (P <0.05). Conclusion DHA can reduce the inflammatory response by reducing the expression of NF-κB, TNF-α and IL-6 and play a role in the treatment of radiation pneumonitis.
