目的研究人参皂苷Rg2对自发性高血压大鼠(SHR)血压的影响及肾脏组织中基质金属蛋白酶(MMP)2和MMP9表达的影响。方法采用无创大鼠鼠尾测压仪测量各组大鼠动脉血压及心率,Realtime PCR检测大鼠肾脏组织MMP2、MMP9的mRNA水平,Western Blot检测MMP2、MMP9的蛋白水平。结果与WKY大鼠相比,SHR大鼠的体质量、肾脏质量、心率均无明显变化,血压包括收缩压、舒张压及平均动脉压均显著性增高(P<0.05)。Rg2处理4周后,SHR大鼠的血压均显著性降低(P<0.05),WKY大鼠的血压无明显变化。SHR大鼠的MMP2和MMP9的mRNA水平均明显高于WKY大鼠(P<0.05)。Rg2处理4周后,SHR大鼠的MMP2和MMP9的mRNA水平均显著下降(P<0.05),WKY大鼠的MMP2和MMP9的mRNA水平无明显变化。Rg2处理后SHR大鼠的MMP2和MMP9蛋白变化趋势与mRNA变化趋势一致。结论 Rg2显著降低SHR大鼠的血压以及肾脏组织MMP2和MMP9的表达。
Objective To study the effect of ginsenoside Rg2 on blood pressure in spontaneously hypertensive rats (SHR) and the expression of matrix metalloproteinase (MMP) 2 and MMP9 in kidney. Methods Arterial blood pressure and heart rate were measured by noninvasive rat caudal pressure manometer. The mRNA levels of MMP2 and MMP9 in rat kidney were detected by Realtime PCR. The protein levels of MMP2 and MMP9 were detected by Western Blot. Results Compared with WKY rats, there was no significant change in body weight, kidney mass and heart rate in SHR rats. Blood pressure including systolic pressure, diastolic pressure and mean arterial pressure were significantly increased (P <0.05). After 4 weeks of Rg2 treatment, the blood pressure of SHR rats decreased significantly (P <0.05), while the blood pressure of WKY rats did not change significantly. The mRNA levels of MMP2 and MMP9 in SHR rats were significantly higher than those in WKY rats (P <0.05). After Rg2 treatment for 4 weeks, the mRNA levels of MMP2 and MMP9 in SHR rats were significantly decreased (P <0.05), while the mRNA levels of MMP2 and MMP9 in WKY rats did not change significantly. Rg2 treatment SHR rat MMP2 and MMP9 protein trends and mRNA trends consistent. Conclusion Rg2 significantly reduces the blood pressure and the expression of MMP2 and MMP9 in kidney of SHR rats.
