目的探讨CXCR4siRNA对人食管鳞癌裸鼠移植瘤生长的抑制作用。方法建立人食管鳞癌细胞株EC9706裸鼠移植瘤动物模型,瘤旁注射化学合成CXCR4siRNA,同时以瘤旁注射阴性对照siRNA和PBS为对照。监测肿瘤生长变化,测量瘤质量。HE染色观察肿瘤病理学变化,RT-PCR、免疫组化方法检测CXCR4mRNA和蛋白变化。结果裸鼠体内实验结果显示,与对照组比较,CXCR4siRNA组肿瘤生长受到明显抑制,瘤质量下降;HE染色显示对照组肿瘤体积大、异形明显;RT-PCR、免疫组化结果表明CXCR4siRNA处理组CXCR4mRNA和蛋白表达下调。结论CXCR4siRNA能抑制人食管鳞癌EC9706细胞株裸鼠皮下移植瘤的形成,且能在体内有效下调CXCR4mRNA和蛋白的表达。
Objective To investigate the inhibitory effect of CXCR4 siRNA on the growth of human esophageal squamous carcinoma xenografts in nude mice. Methods The human esophageal squamous carcinoma cell line EC9706 xenografts in nude mice was established and the CXCR4 siRNA was synthesized by tumor injection. At the same time, the tumor-negative control siRNA and PBS were used as controls. Tumor growth was monitored to measure tumor mass. The pathological changes of tumor were observed by HE staining. The changes of CXCR4 mRNA and protein were detected by RT-PCR and immunohistochemistry. Results The results of in vivo experiments in nude mice showed that the tumor growth of CXCR4 siRNA group was significantly inhibited and the tumor mass was decreased compared with that of the control group. The HE staining showed that the tumor volume was large and the abnormal shape was obvious in the control group. The results of RT-PCR and immunohistochemistry showed that CXCR4 mRNA And protein downregulation. Conclusion CXCR4 siRNA can inhibit the growth of human esophageal squamous carcinoma cell line EC9706 in nude mice and effectively down-regulate the expression of CXCR4 mRNA and protein in vivo.
